Outline of Research at the Start

骨の成長には軟骨から骨に変化する軟骨内骨化と、直接骨を形成する膜性骨化に大きく別れる。顔面の成長は上顎骨と下顎骨の前方への成長により行われるが、下顎骨の成長では関節頭の関節軟骨における軟骨内骨化と膜性骨化が関与し、上顎骨の成長でも膜性骨化だけでなく、その付近に存在する蝶形骨や篩骨、後頭骨の間の軟骨内骨化が深く関係することが知られている。このような顔面の発育に関係する軟骨内骨化について、自然発症型の軟骨内骨化遅延(Cartilage Calcification Insufficient, CCI)ラットを対象にその原因遺伝子や軟骨内骨化に異常をきたすシグナルについて解析する。

Outline of Final Research Achievements

The relationship between gene mutation and developmental delay of cartilage calcification insufficient (CCI) rat have been identified. This rats showed an autosomal recessive genetic disorder, similar to rickets/osteomalacia in humans, and revealed that the endochondral ossification delay of the cranial base and mandibular condyle cartilage, were associated with the over expression of Gli1 mRNA, a transcription factor of the Indian hedgehog (IHH) signalling pathway, which resulted to increased proliferation of chondrocytes and impaired differentiation.

Academic Significance and Societal Importance of the Research Achievements

本研究は、骨格の発育異常としてみられるヒトのくる病に極めて類似した症状を示すことから、原因遺伝子の解明や発現する分子間シグナル伝達の均衡の乱れが考えられました。検出された遺伝子は軟骨細胞の増殖と分化を司る重要な因子の一つであるGli1の過剰発現によることがわかりました。これらの発育異常ラットの病態解明は将来的な分子標的治療薬開発の指標となり得る点で、病態の原因解明が治療法開発に繋がることが期待されます。
