Outline of Research at the Start

申請者らは、マウスの TAC 処理下において Dec1 が心臓線維化を促進することを新たに示した (Int J Mol Sci 2019)。Dec1 が TNF-α や Smad3 などを促進させることを示した。さらに、Dec1KO (ノックアウト) マウスは、血管周囲線維化から心臓を保護し、M1/M2 マクロファージ分極を調節し、心不全に伴うアポトーシスを減少させることを示した。これらのDNAマイクロアレイ解析により、APEX1、WNT16、FGF10、MMP-10 遺伝子が発現され、マイクロRNA (miRNA) アレイ解析では、対応する miRNA (miR-295、miR-200b、miR-130a、miR-92a) は同じ傾向を示した (Biochem Biophys Res Commun 2020)。

Outline of Annual Research Achievements

miRNA 21の標的遺伝子Dec1であることを特定し、in vivo研究では、miRNA21のノックアウトマウスを作成した。マウス歯周病モデルにおいて、生後3ヶ月齢のC57BL/6マウスおよびmiRNA21ノックアウトマウスの歯槽骨吸収が正常マウスのそれよりも有意に低いことを発見した。 さらに、miRNA21 の欠如が骨芽細胞の骨形成分化機能を大幅に低下させることが示されており、M-CSF および RANKL 誘導骨髄細胞では、酒石酸染色陽性の破骨細胞の数も大幅に減少しており、miRNA21 が欠如していることが示唆された。miRNA21が欠乏すると、骨芽細胞の骨形成分化機能が著しく低下し、骨芽細胞や破骨細胞の機能にも影響を及ぼした。パラフィン切片では、破骨細胞の数が大幅に減少し、miRNA21 ノックアウトマウスの正常な機能は、主に破骨細胞の機能不全によるものであると推測された。 一方で、miRNA21 欠失は、関連遺伝子またはタンパク質の発現の変化を引き起こし、破骨細胞の分化に直接影響した。C57BL/6マウスと miRNA21 ノックアウトマウスの歯周病前後における実験として、OPG/RANKL/RANK 軸の変化パターンを検証したところ、C57BL/6マウスに比べて miRNA21 の血清中の RANKL 値が上昇していることが判明した。

Reason

計画書に沿った進行を行っているが、miRNA 21の標的遺伝子Dec1であることを特定し、in vivo研究では、miRNA21のノックアウトマウスの作成やその系の立ち上げ等に時間を要しているため、計画書以上の進展はない。これら問題点は、これまでの各種検討により徐々に解決されていくと考えられるため、今後は更なる研究の進展が期待される。また、並行して進めているmiRNA21のノックアウトマウスのTACモデルの作成やその系の立ち上げ等に時間を要しているが、こちらもこれまで検討を基に、今後研究が進展すると期待されている。
以上のことから、現在までの進捗状況は、計画書を大きく超えて進展するものではないものの、おおむね順調に進展しているものと判断している。

Strategy for Future Research Activity

歯周病モデルを用いて TAC にて同定された miRNA 機構の解明:
A) 実験的歯周病モデルマウスを用いて、TAC により同定された miRNA の in vitro 解析 (リアルタイムPCR法) を行う。
B) 血漿の解析 (ELISA法)、顎骨の in vivo 解析 (マイクロCT、ヘマトキシリン・エオジン染色法、免疫染色および TRAP染色法) にて検討する。
