| Outline of Research at the Start |

| Outline of Final Research Achievements |
There are variations in the number of digits between species. However, the mechanism that explains the morphological diversity of digits was unclear. In this study, we conducted a genetic analysis of genes related to digit development in the urodele amphibian newt (Pleurodeles waltl). To reveal gene functions for the digit development in newts, we produced knockout newts using CRISPR/Cas9. Gli3 crispants (KO) newts show anterior digit duplication phenotype in fore- and hindlimbs. Tbx3 KO newts show posterior digit deletion in forelimbs. These phenotypes were similar to those of the KO mice and human genetic syndrome associated with each gene. However, partial differences in gene function appeared. Tbx3 KO newt hindlimb shows one digit addition. Further research shows that different combinations of Tbx family genes can affect the number of digits and alter Shh expression in the limb. This research offers valuable insights into the evolution of diverse digit morphologies.
