Outline of Research at the Start

自閉スペクトラム症や注意欠陥・多動性障害をはじめとした神経発達症の患者数は非常に多いことが知られるが、その発症機構は遺伝的素因と環境要因が関与することが知られている程度であり、詳細は不明である。申請者は、発生期の神経幹・前駆細胞のニューロンサブタイプ分化能変化に重要な役割をもつ転写因子のひとつとしてPHF21Bを見出した。この因子はその構造から、エピゲノム制御に関与することが予想されるが、その役割は不明である。そこで、通常よりも迅速に行える方法により、Phf21bの遺伝子改変マウスを作製し、エピゲノム制御の観点から発症機構に関与するか否かを明らかにする。

Outline of Final Research Achievements

We identified that three transcription factors, including Phf21b, contributed to a spatiotemporal differentiation potential of neural stem/progenitor cells (NSPCs) into neuronal subtypes in vivo. In cortical development, overexpression of these three factors by the in-utero electroporation (IUE) at mouse E13.5 revealed an increased proportion of lower-layer neurons in the late development stage (E17.5). Moreover, we found that transcription or translation activity changes may be involved in this regulation.

Academic Significance and Societal Importance of the Research Achievements

初期型ニューロンサブタイプ分化能の責任因子の一つとして同定したPHF21Bは、そのタンパク質ドメインからエピジェネティクス制御に関与すること、さらに、発語・精神遅滞、発達障害を示すPhelan-McDermid症候群(22q13.3欠失症候群)関連染色体上に位置することから、神経発達症や大うつ病性障害等の精神疾患に関与している可能性が考えられるが、その役割は不明である。Phf21bの機能阻害あるいは遺伝子改変マウスの表現型解析は、神経発達症の新規モデルマウスの開発、将来的には病態解明、新規治療法の開発につながることが期待できる。
