Outline of Research at the Start

糖尿病腎症への対策は医療上・医療経済上の重要課題であるが、いまだに糖尿病が腎不全を引き起こすメカニズムには未解明の点が多い。糖尿病にさらされた腎臓では様々なストレス応答機構が働いており、それらがお互いに影響を及ぼし合って病態を形成していると考えられる。本研究では、小胞体とミトコンドリアの相互作用に着目して解析することで糖尿病腎症の本質的な治療標的の同定を目指す。

Outline of Annual Research Achievements

腎臓病の進行過程において、糸球体足細胞(ポドサイト)には様々なオルガネラストレスが同時に生じているが、オルガネラ同士の連携についての検討は不十分である。小胞体膜に存在するPDZ domain-containing 8 (Pdzd8) は複数のオルガネラ繋留を担うことが報告されている。本研究では、ポドサイト特異的にPdzd8を欠失させたマウスを用いて、ポドサイト障害におけるオルガネラ連関の役割を解明することを目的とした。
まず、Pdzd8をポドサイト特異的にノックアウトしたマウスに肥満関連腎症モデルを作製し、病態解析を行った。ポドサイトでのPdzd8欠失は肥満関連腎症モデルでの尿中アルブミンを増加させ、ポドサイト内に脂肪分に満ちた異常エンドソームの蓄積を招いた。単離糸球体のプロテオーム解析では、Pdzd8欠失によってミトコンドリア・エンドソーム恒常性に関わるタンパク質が有意に変動することが示された。
これらの機序を調べるため、培養ポドサイト細胞を用いた実験を行った。培養ポドサイト細胞では、Pdzd8ノックダウンによってミトコンドリア機能が低下し、小胞体とミトコンドリアの接触が減少していた。さらに動物実験と似た異常エンドソームの蓄積も確認された。リピドーム解析では、Pdzd8ノックダウンによってポドサイト内にグルコシルセラミドの蓄積が認められ、異常エンドソーム蓄積との関連が示唆された。
これらの結果から、Pdzd8を介したオルガネラ連関は脂肪毒性にさらされたポドサイトの恒常性維持に働くことが明らかになった。
