Outline of Research at the Start |
悪性黒色腫において、大規模なゲノム解析の報告はあるものの、エピゲノム・ゲノム関連に注目した解析はいまだ十分でない。そこで、本研究では日本人を対象とした悪性黒色腫のゲノム、エピゲノム変異を解析し、新たな治療ターゲットを解明することを目的とする。日本人悪性黒色腫の臨床検体を、DNAメチル化に注目する独自性の高い解析を行い、メチル化を介して腫瘍形成に関わる、新たな治療ターゲットとなりうる遺伝子とその機能を解明する。

Outline of Final Research Achievements |
In clinical samples of patients with malignant melanoma, the group of patients was stratified by focusing on aberrant DNA methylation of genes and correlated with clinical symptoms. Genetic abnormalities correlating with these stratifications were also identified. Functional analysis of malignant melanoma cell lines revealed that knockdown of certain genes increased cell proliferation and invasiveness. These findings suggest that the expression of specific genes may be decreased due to abnormal DNA methylation, which may be involved in tumor growth.

Academic Significance and Societal Importance of the Research Achievements |
悪性黒色腫に対する化学療法は免疫チェックポイント阻害薬と分子標的薬が上市されているが、免疫チェックポイント阻害薬の奏効率は約44%であり、分子標的薬に関しては適応となる患者が日本人では25%とまだこれら薬剤の恩恵にあずかれない患者も多い。 我々の研究では腫瘍のエピゲノム情報に注目することにより、新たな治療対象となりうる遺伝子を検索した。その結果いくつかの候補遺伝子が得られ、機能解析にて表現型に寄与しうる遺伝子を同定することができた。
