Outline of Research at the Start

皮膚は、身体の外と内を分かつ境界であり、「バリア」として身体の恒常性の維持に貢献している。皮膚バリアの破壊に伴うドライスキンは、アトピー性皮膚炎(AD)や乾癬等の皮膚炎を引き起こす。
一方、制御性T細胞(Treg)は過剰な免疫応答を抑制する役割を担っている。Tregの異常により引き起こされるIPEX症候群では、皮膚炎が認められ、Tregが皮膚の恒常性の維持に関与していることが示唆されるが、その詳細なメカニズム及びTregと皮膚バリアとの関係性は不明である。そこで本研究では、皮膚バリアにおけるTregの役割の詳細を解明し、ADを代表とする炎症性皮膚疾患の新規治療法及び予防法の開発を目指す。

Outline of Annual Research Achievements

ヒトの身体の最も外側に位置する皮膚には、物質の透過を防ぐ強力なバリアが存在している。この皮膚バリアは病原体、抗原、有害物質の侵入を阻止し、体内からの水分の蒸発も防いでいる。皮膚バリアの破壊に伴うドライスキンは、アトピー性皮膚炎(AD)や乾癬等の皮膚炎を引き起こす。
一方、免疫系の恒常性維持に働いているT細胞である制御性T細胞(Treg)のマスター転写因子であるFoxp3の変異や欠損より引き起こされるIPEX症候群やscurfyマウスでは、しばしば皮膚炎が認められ、皮膚恒常性にTregが関与することが示唆されるが、皮膚バリア機能とTregの関係性は不明瞭である。
そこで本研究は、ADを代表とする炎症性皮膚疾患の新規治療法及び予防法の開発を目指し、皮膚バリア破綻におけるTregの役割を解明することを目的として、皮膚バリア破綻モデルマウス(4% SDS塗布)の解析を行った。その結果、皮膚バリア破壊によってTregが真皮内に浸潤し、抑制性サイトカインを産生していることを見出した。また、Tregの浸潤には表皮角化細胞から放出されるIL-33が直接関与していることを発見した。更なる解析の結果、 ADモデルマウス(4% SDS + ダニ虫体成分含有軟膏塗布)では、Treg数がコントロール群である皮膚バリア破壊モデルマウスに比べて減少することを見出した。現在なぜADモデルマウスではTreg数が減少するのかを、ダニ抗原由来プロテアーゼに着目して研究している。
皮膚恒常性及びAD病態形成機序の解明は炎症性皮膚疾患の新規治療法及び予防法の開発に繋がる。

AD発症メカニズムにダニ抗原由来セリンプロテアーゼ‐IL-33-Treg軸が関与していることを示唆する結果が得られたため。

Strategy for Future Research Activity

これまでに行った実験の結果、AD発症メカニズムに抗原由来セリンプロテアーゼが関与し、IL-33-Treg軸を揺るがすことが判明した。来年度はIL-33-Treg軸を揺るがす抗原由来セリンプロテアーゼの同定を行い、AD発症メカニズムの解明及び新規治療法の開発に繋げたい。
