Outline of Research at the Start

FIB-SEMを用いた核内クロマチン構造の3次元電子顕微鏡像観測，基準点となるスピンドル極体(SPB)と標的とする遺伝子座に蛍光タンパク質で標識導入した分裂酵母を対象に，核内での遺伝子座の動態計測を共焦点顕微鏡で行う．2つの計測データをもとに，粗視化モデルとして核内クロマチン構造・動態を表現する数理モデルを構築する．構築する数理モデルを用いて，核内クロマチン構造・動態をシミュレートすることにより，核内クロマチン高次構造の遺伝情報制御における役割を解明する．

Outline of Annual Research Achievements

本年度は，3Ｄ電子顕微鏡像解析のために必要となるクロマチン構造動態モデルの精密化を中心に進めた．もともと計画していた，遺伝子座に導入した約130箇所の蛍光標識点間距離の変化データをもとに構築するクロマチン構造動態モデルでは，3Ｄ電験像を一義的に再現するには曖昧さが大きすぎることが明らかになった．これは当初から想定していた懸念事項であった．不足する構造情報を補い，なおかつクロマチン構造の動態を再現するためのモデルの導入が必要であるため，理研・新海博士が開発したモデル技術（Phi-C)の導入を今期行った．
Phi-Cは，Hi-Cデータをもとにして，遺伝子座間の核内での接触頻度を引力・斥力の仮想的なポテンシャルに還元する．さらに，この仮想的なポテンシャルを用いることでクロマチン構造の動的構造変化のシミュレーションを可能とする．
私達の新しいアプローチでは，Phi-Cモデルで得られる仮想ポテンシャルに加えて，実験的に観測された130箇所の蛍光標識点の動態，蛍光標識点間の距離の時間変動をさらに構造制約として加えることで，動的な実験情報を加えたより性格なクロマチン構造の動的モデルを構築する．新たなクロマチン構造動態モデルを基にしてクロマチンの3Ｄ電子顕微鏡像の解析を進める．
共焦点顕微鏡で取得した時分割データについても，誤差の評価をやり直し，より精度の高い標識点間の距離情報の取得が可能となるように画像データ解析法を改善した．
電子顕微鏡像取得については，引き続き染色法を改善したデータの習得を継続しており，最終的に構造解析するためのより良い電子顕微鏡像の取得を平行してい進めている．

Reason

当初計画していたクロマチン構造動態モデルの限界が明らかになり，構造動態モデルの高度化が必要になった．ただ，当初より実験情報の数には制約があるので，異なる構造情報の導入の必要性は考えながら進めていたので，想定内の計画変更である．
理研・新海博士のPhi-Cモデルについは私達の実験データを導入することで，より実験情報（特に動態に関する情報）を取り入れた精密なクロマチン構造動態モデルへの展開が可能であると考えていた．新海博士と議論を重ねて，私達のクロマチン動態データをも構造制約として取り入れたモデルの高度化に向けた共同研究を始めた．
分裂酵母のHi-Cデータは複数公開されているので，公開データを用いて現在モデルの高度化を進めている．

Strategy for Future Research Activity

クロマチン構造動態モデルの高度化を進める必要性が明らかになり，新たにHi-Cデータにもとづいてクロマチン構造動態情報を取得するPhi-C法を導入することにした．
研究の方向性や最終目標に変更はないが，当初のモデルでは電子顕微鏡像解析を進めるには解析精度に限界があるために，新たな構造解析技術の導入が必要であった．
Hi-Cの公開データを使い，すでにモデル構築を進めている．
全体の計画に大きな遅れをもたらすことなく研究を進められる．
