Outline of Research at the Start

FIB-SEMを用いた核内クロマチン構造の3次元電子顕微鏡像観測,基準点となるスピンドル極体(SPB)と標的とする遺伝子座に蛍光タンパク質で標識導入した分裂酵母を対象に,核内での遺伝子座の動態計測を共焦点顕微鏡で行う.2つの計測データをもとに,粗視化モデルとして核内クロマチン構造・動態を表現する数理モデルを構築する.構築する数理モデルを用いて,核内クロマチン構造・動態をシミュレートすることにより,核内クロマチン高次構造の遺伝情報制御における役割を解明する.

Outline of Annual Research Achievements

2024年度は、クロマチン構造動態モデルを最適化してモデルとして完成とともに,そのモデルから取り出せる構造・動態情報の検証を集中的に進めた.
理研・新海博士の協力を得て,彼が開発したPHi-Cを元にして131カ所の遺伝子座の動態データと,Hi-Cから得られている遺伝子座間の近接データを元にして,遺伝子座間に仮想的なポテンシャルを構築することで,分裂酵母の内の3つの染色体の立体構造・動態シミュレーションを可能とした.
今期は,各クロマチン構造・動態シミュレーションを詳細に解析することで,その立体構造・動態の生物学的な意味について解析を進めた.シミュレーションデータから,各染色体のセントロメア,テロメア間の距離の分布を解析したところ,実験データと高い整合性が確認された.モデルの妥当性が確認できた.
分裂酵母のセントロメアはSPBと呼ばれる一カ所に集積しているが,SPBは周期的な揺動運動をしており,その結果クロマチン構造全体が大きく揺動することが予想されていたが,今回のPHi-Cモデルによる解析では,セントロメア周辺は大きく揺動するも,セントロメアから離れるにつれて,クロマチン構造の運動は急激に低下するという特徴が明らかになった.クロマチン構造は核内では意外なほど同じ形態を維持しているという意外性が見いだされた.
このことは,クロマチン構造の電子顕微鏡像再構築の際には,むしろ有利に働く.今期の成果をもとに,FIB-SEM像の再構築へと研究を進める.

3: Progress in research has been slightly delayed.

Reason

クロマチン構造・動態モデルは当初予定してよりも格段に精度の高い完成度の高いものにすることができた.一方で,2024年度中は,研究代表者の家庭で家人が逝去、別の家族が入院治療するなどのため、代表者の研究時間が分断されたため,モデル構造からすぐに電子顕微鏡像解析という最終ゴールに到達する時間が十分にとれない事情があった.このため,一年間研究期間を延長して,最終的なゴールまで研究を進める.
2024年度の解析により,当初予想していたのとは異なり,分裂酵母の核内クロマチン構造は意外なほど大きな構造変化をしておらず,むしろ一定の立体構造を保つという描像が得られたのは電子顕微鏡像解析を行う上では,構造多形を仮定した解析が必要なくなったために,計画よりもスムーズに進めることができることがわかった.

Strategy for Future Research Activity

2024年度は,研究代表者の私的な事情もあり,クロマチン構造・動態モデルに基づいた電子顕微鏡像解析という最終ゴールへの到達ができなかったが,一年の延長期間のうちに確実に電子顕微鏡像解析まで進める.
これまでの段階で,想定した以上に高精度なクロマチン構造・動態モデルの構築ができたことは強調したい.膨大な独自の実験データに基づいて,オリジナルな統計物理モデルによる独自のクロマチン構造モデルである.現在,本研究で開発したクロマチン構造・動態モデルの部分の論文を作成しており,2025年中には論文発表する予定である.
