Outline of Research at the Start

1) 中枢神経細胞特異的に蛍光標識tetraspaninを発現するマウスを作製する
2) 中枢神経細胞由来のexosome特異的なマーカーを確立する
3) 2)によって得られた中枢神経細胞特異的exosomeマーカーを利用して末梢血より神経細胞由来のexosomeを精製する方法を確立する

Outline of Final Research Achievements

Liquid biopsy is an extremely useful method for disease diagnosis; however, it is difficult to perform liquid biopsy in peripheral blood because cerebrospinal fluid must be collected for CNS diseases. In this study, we aimed to establish a reliable CNS-derived exosome marker in peripheral blood. 1) We generated mice expressing fluorescently labeled tetraspanin specifically in CNS. 2) We established a CNS-derived exosome-specific marker. 3) We established a method to purify the exosomes derived from neurons from peripheral blood using the CNS-specific exosome marker obtained by 2).

Academic Significance and Societal Importance of the Research Achievements

中枢神経疾患は生前において病理学的な確定診断を得ることがほぼ不可 能であり,末梢血liquid biopsyによるバイオマーカー診断が重要な役割を果たすポテンシ ャルを有する.Exosomeは異なる細胞間の情報伝達に重要で,由来する細胞の情報を保有す ることから,癌においては発症や転移のマーカーとして注目されている.神経においても Exosomeが病態において重要であるとする報告はあるものの,現在提唱されている中枢神経 由来のexosomeマーカーの信頼性は著しく低い.本研究では,信頼性の高い中枢神経細胞 由来exosomeマーカーを確立できた。
