Outline of Research at the Start

健康寿命延伸の実現には,骨格筋老化(加齢性筋減少症)の防止は重要な課題である. これまでの研究成果から,骨格筋老化は,骨格筋の微小環境形成の不均衡による増殖・再生能低下が原因の一つと考えられた.本研究課題では骨格筋の老化を防止するために,微小環境の安定性の維持や,骨格筋の老化とともに起こる幹細胞の自己複製能や筋量の低下の防止に関わるエクソソームを解析し,骨格筋の維持・再生における動植物由来成分によるエクソソームの影響を検証する.

Outline of Annual Research Achievements

健康寿命の延伸の実現には,骨格筋老化(加齢性筋減少症)の防止は重要な課題である。これまでの研究成果から,骨格筋老化は,骨格筋の微小環境形成の不均衡による増殖・再生能低下が原因の一つと考えられた。本研究課題では骨格筋の老化を予防するために,微小環境の安定性の維持や,骨格筋の老化とともに起こる幹細胞の自己複製能や筋量の低下の防止に関わるエクソソームを解析し,骨格筋の維持・再生における動植物由来成分によるエクソソームの影響を検証することを目的とした。
骨格筋細胞を用いて食品等動植物由来成分を添加しエクソソームを放出する成分を,その量とmiRNAの解析によって機能性を有する候補物質をスクリーニングすることを目的とした。このために,食品等動植物由来成分を探索し,抽出や精製を行い,細胞に添加し細胞への影響の観察を経時的に行ったところ,複数の食品が候補としてスクリーニングされた。また,これらの抽出成分を添加した細胞の解析と,添加至適濃度を決めるための条件設定実験を行った。また,エクソソームの解析について実験条件の設定を検討した。食品等動植物由来成分については,多種類のサンプル収集や測定実験においてコロナウィルス予防対策に関連する制限があったため,複数年度において食品等動植物由来成分の種類を追加し,筋線維への添加実験を行っている。実験動物にこれらの動植物由来の機能性成分を摂取させ,成分によって誘発されたエクソソームへの影響の解析や,含有するmiRNAや発現タンパク質の解析を実施し,筋再生の促進に対する影響について検証を行っている。

細胞実験において対面の実験の整備と,機材整備を含め効率化をはかり,サンプルの収集を行い研究を推進する計画である。
食品等動植物由来成分による影響を評価するために,エクソソームの抽出・解析,miRNAの解析,ターゲットタンパク質の解析を行い,さらに筋再生ニッチ形成と微小循環の関連性を検証するため微小血管構造の解析を実施し,実験動物での骨格筋再生について検証する方策である.
