Outline of Research at the Start

近年我々は優れたワクチン候補分子であるRipr5を見出すことに成功し、赤血球気マラリアワクチンとしての実現を目指している。しかしコロナウイルスワクチンと同様にRipr遺伝子の点変異がワクチン効果に及ぼす影響は無視できない。そこで動物に免疫して得られた抗Ripr5抗体を用いて、ケニアで流行しているマラリア臨床分離株を対象に原虫増殖阻害試験を行う。また同時に、Ripr遺伝子配列を決定し、遺伝子変異と原虫増殖阻害試験の結果を比較することによって、Ripr5のワクチン効果がRipr遺伝子の変異によって影響を受けるかどうかを試験する。以上によりマラリアワクチンの実現を強力に推進することができる。

Outline of Annual Research Achievements

我々は逆ワクチン学的アプローチによって、新規なマラリア赤血球期ワクチン候補抗原であるRiprを見出した。しかしその構造と配列は非常に複雑かつ巨大であり、特に分子内にシステイン残基を87個も有するなど、ワクチン開発に困難であると考えられた。そこで更にRipr分子を11個の領域に切断し、同様な逆ワクチン学アプローチを行うことによって、PfRipr5を見出した。配列は非常に保存されているが、現在まで得られた情報は標準株に対する増殖阻害活性のみである点が課題として残されている。そこで本研究はケニアで得られた患者血液を用いて原虫をex vivoで抗Ripr抗体で増殖阻害活性を測定することにより、実際の流行株でもRiprが有効である事を検証するものである。
現在までに、複数の現地由来株の全ゲノム解析を行い、Ripr遺伝子配列の保存状態を確認した。その結果、他の地域と同様に高度に保存されている事がわかった。また本研究に必要なIRBの申請を行い、問題なく受理された。ex vivoに用いる抗体はすでに準備されており、標準株に対する増殖阻害活性の確認を実施済みである。本年度はビクトリア湖周辺・ビクトリア湖にある島(ムファンガノ島)にてサンプルを採取し、すぐにex vivoの増殖阻害活性を測定する実験を行うことができた。また本研究センターと相手大学との間でMOUを締結することに成功し、ABS対応を万全のものとした。

Reason

実際にマラリア患者から血液を採取し、そのままフィールドでの培養方法の確立に成功した。Ripr5抗体による増殖阻害活性を測定することに成功した。マラリア患者数は非常に多く、一日に20例を数えるほどであり、予想よりも効率的に実験を遂行できると考えられる。また愛媛大学プロテオサイエンスセンターとマウントケニア大学間でMOUの締結に漕ぎ着けた。

Strategy for Future Research Activity

MOU締結に成功したので、懸念事項の一つであった、フィールドアイソレートの遺伝情報のABSに関するイシューが解決された。そこでRipr5遺伝子における多型とRipr5抗体による増殖阻害活性との相関を明らかにする基盤が整った。本年度は積極的にフィールドに赴き、ex vivo実験を重ねる予定である。
