Outline of Research at the Start

歯周病の病態形成における OMV-sRNA の意義と分子機構の解明を目指し、OMV-sRNA を標的とした歯周病の診断や治療薬の開発を展望する。

Outline of Annual Research Achievements

Porphyromonas gingivalis (Pg)の細胞、およびOMVのRNA-Seq解析を行った。その結果、細胞内・OMV内に蓄積する複数の新規small RNAを同定した。経時的なRNAの発現量の推移について、ノーザンブロットとReal-Time PCRにて解析を行った。その結果、時期特異的発現を示すものや、転写後プロセシングを受けるものがあることが明らかとなった。
また、Pgの形質転換法を新たに開発した。この新規の形質転換法は、これまでの電気穿孔法や接合伝達法と比較して、高い効率で形質転換することが可能であった。上記の新規small RNA遺伝子欠損株も当該手法により容易に作成することができた。
Pg野生株とIX型分泌機構 (T9SS) 欠損株の菌体及びOMVsを用いて、それらの微細構造と病原因子の局在を可視化できる凍結割断/走査型顕微鏡(SEM) 法の確立に取り組んだ。野生株の菌体の割断面と非割断面に対するA-LPS抗体と金コロイド二次抗体による免疫SEMでは、割断面の菌体外周と非割断面の菌体表層にシグナルを得た。

Pgの凍結割断法が間もなく確立できる見込みであり、sRNAの研究で成果があり、学会発表などを行った。

Strategy for Future Research Activity

細胞内・OMV内の主要な新規small RNAの役割を明らかにするために、in silico解析によるターゲットmRNA予測とwet実験による検証、smallRNA遺伝子欠損株を用いたtranscriptomeやproteome解析、新規small RNA遺伝子の過剰発現系、発現・局在レポーター系の構築等を行う。
細菌及び細菌付属器、膜小胞などの微細構造を観察する凍結割断/走査型顕微鏡(SEM) 法を確立する。
