Research Abstract

本研究は、研究代表者がこれまで研究してきた機能抗体断片を構成要素として接合対象素材によらず目的素材間をナノレベルで正確に接合・アセンブリ制御できるバイオインターフェイス分子を創出するプラットフォーム技術を開発し、様々な生体分子・細胞・工学ナノ材料に応じたインターフェイス分子の迅速な適応・進化を可能にする接合分子デザイン工学を提言することを目的とする。そのために、本年度は、下記の研究を行った。
1.タンパク質フォーマットを利用した組換えバイオインターフェイス分子の構築
異なった機能を有する複数の免疫グロブリンフォールドを機能・構造単位と捕らえ、抗体分子骨格を母体として高次多重特異性を持つ組換えバイオインターフェイス分子を、X線結晶構造解析情報を基に作製するため、抗体の抗原結合ドメインのみをペプチドリンカーで融合した二重特異性抗体を大腸菌発現系で調製し、リンカーの設計によってタンパク質の発現量が変化することが分かった。
2.組換えバイオインターフェイス分子の構造・機能解析
1.より作製したバイオインターフェイス分子の構造解析をX線結晶解析などによって行う検討を始めた。細胞架橋用バイオインターフェイス分子については、単独結晶化による構造解析へ向けたサンプル調製の最適化のプロセスを設計した。また、ナノ工学材料特異的抗体断片に関しては、電子顕微鏡によるバイオインターフェイス分子による観察を行うために最適なサンプル調製を見出した。
