ダウン症候群因子DSCR-1の複合体、生理機能解明:血管からの抗がん活性応用

Research Abstract

腫瘍血管内皮増殖に寄与する増殖因子(VEGF)やトロンビン刺激での内皮遺伝子変動の網羅的解析から刺激早期に共通して最大に誘導されるダウン症候群関連因子(DSCR)-1を見出し、この因子が内皮活性化の重要転写因子NFATのnegative feedback因子として作用すること、DSCR-1の発現を促すことで血管新生、腫瘍増大が顕著に抑制されること、急性炎症状態における心肺機能低下を抑制し、マウス敗血症での致死率を大きく低下させることを明らかにしてきた。今年度は、NFAT/DSCR-1 feedback signalにおける内皮機能への効能をさらに分子レベルで解明するため、内皮で発現するNEAT、ファミリーの一つNFATc1の特異的モノクロナール抗体の作製から、NFATc1によるクロマチン免疫沈降一次世代シーケンサー(ChIP-seq)を行い、ゲノムワイドなNEATc1の内皮細胞結合プロファイルを作製することに着手した。その結果、NFATc1抗体は高感度で特異的な反応を示し、かつ臍帯静脈内皮細胞(HUVEC)を用いたChIP-seqにおいてNEATc1は定常状態でも有意な結合を特定部位で示すこと、しかしVEGF刺激に伴い、このNFATc1の結合量は大きく増大し、DSCR-1のみならず、腫瘍増大に関連するAng-2やダウン症相関因子DYRK1Aのpromoter上にも結合しうることを見出した。
