Research Abstract

この研究は、虚血・再灌流の際、心筋細胞内Na・Ca濃度と活動電位変化を予測することを目指す。そのため、22年度のミトコンドリア酸化的リン酸化モデルの数核的解析、ATP/ADP濃度の変化に最も大きな依存性を持っているNa+/K+ATPaseの新しいモデルの開発に続け、ヒト心筋細胞モデルの開発、膵臓β細胞モデルの開発を行った。
1.ヒト心筋細胞モデルの開発とミトコンドリア酸化的リン酸化モデルの導入。
虚血・再灌流の影響を正確に予測することを目指し、ミトコンドリア酸化的リン酸化モデルをヒト心筋細胞モデルに加えた。去年行った、酸化的リン酸化モデルの数学的な解析(bifurcation analysis)の結果を元にparameterの再調整を行い、正常の広い範囲の心臓拍動頻度に対して細胞の機能が正常に働くことを成功的にシミュレーションできた。それに加え、去年発表した新しいNa+/K+ATPaseモデルを心筋細胞モデルに導入した。このモデルは虚血・再灌流の際、細胞外の酸素・グルコースの濃度などが変わる時に細胞のATP/ADP変化を予測するのに基本ベースになると期待できる。
2.膵臓β細胞モデルの完成とNa・Ca濃度と活動電位変化の数学的解析
ATP/ADP濃度の変化によるNa・Ca濃度と活動電位変化に最も大きな影響を与えるのは、Na+/K+ATPase
activityの変化である。去年発表した新しいNa+/K+ATPaseモデルをβ細胞に加えることで、細胞外グルコースによって変化する細胞内Na・Ca濃度と活動電位をミュレーションすることができた。その原因になるメカニズムを明らかにするため、我々独自に開発したlead potential analysisとbifurcation analysisを適用し、Na・Ca濃度と活動電位変化に対する細胞各要素の役割を数学的に示すことができた。
