非筋細胞における力学的ホメオスタシスの発現メカニズムの解明

Research Abstract

組織レベルで存在が知られていた力学的ホメオスタシスの分子メカニズムを細胞レベルで明らかにすることが目的である。平滑筋細胞にGFP-nonmuscle-α-actininを発現し、細胞挙動の観察を行った。ミオシン軽鎖のリン酸化は、非筋II型ミオシンの収縮活性に必須であることが知られている。この収縮能は(ミオシン軽鎖の脱リン酸化を促すミオシンフォスファターゼを含め)セリン・スレオニン・フォスファターゼの阻害剤であるカリキュリンAを細胞に投与すると、さらに活性化される。この時のストレスファイバーに沿ったnonmuscle-α-actininの観察から、ストレスファイバーのサルコメア間隔が(擾乱を受けたとしても1時間程度の時間を経て)一定に維持されることが分かった。サルコメア間隔はストレスファイバーが発生する張力と比例関係にあるために、サルコメア感覚が一定に保持されることは張力が一定に保たれることを意味する。すなわち力学的ホメオスタシスとは張力のホメオスタシス(恒常性)に相当することが示唆された。このホメオスタシスの必要条件として、ミオシン軽鎖のリン酸化、およびアクチン重合・脱重合能が挙げられることを確認した。例えばアクチンの重合を阻害するラトランキュリンAにより細胞を処理すると、ストレスファイバーは太くなっていくが、その太さを元に戻す(張力ホメオスタシスを担う)作用ははたらかなかった。また太くなる一方で、ストレスファイバーの発生張力は減少した。これらの一連の現象を矛盾無く説明できる理論モデルを提示するに至った。このように非筋細胞に存在する力学的ホメオスタシスを説明可能な分子・物理モデルが作られ、そのメカニズムの解明に近づいた。今後はこのモデルを様々な実験を通してさらに検証していく予定である。
