Research Abstract

Screening of novel Arabidopsis mutants was carried out to identify ascorbate transport mechanism in plants. Two possibilities have finally obtained among approx. 8, 000 EMS-treated M2 Arabidopsis vtc seeds based on a high-throughput screening by a combination of non-photochemical quenching(NPQ) measurement and histochemical staining with silver nitrate. In addition, based on data from in silico prediction and practical evaluation of ascorbate up-take activity in Arabidopsis T-DNA inserted mutants, it has indicated that at least two genes have a possibility to be the target gene.
