The CpG sites in the Robo4 proximal promoter is specifically non-methylated in endothelial cells(ECs) but not non-ECs. In this study we performed assays using DNA methyltransferase inhibitor, methylated reporter plasmids, and ES cell lines containing mutant promoters, and demonstrated that cell-type-specific DNA methylation of the 11 CpG sites in the proximal promoter regulates EC-specific Robo4 gene expression.
