Research Abstract

唾液腺悪性腫瘍の頻度は全腫瘍の1%程度であり、全身的には比較的珍しい腫瘍である。しかしながら、小唾液腺や舌下腺由来の唾液腺腫瘍は悪性型の占める割合が高く、口腔顎顔面領域における腫瘍学の中で、唾液腺腫瘍の位置付けは非常に高い。また、唾液腺には腺管や腺房を構成する腺上皮細胞や腺房細胞に加えて、筋上皮/基底細胞が存在し、これらが腫瘍化に伴って様々な割合で分化・増殖するため、多様な組織像を示すため、これらの多様さ、不均一さから唾液腺腫瘍を精度高く診断することは困難を極めていることから、より精度の高い診査法が望まれている。
申請者は、多くの組織幹細胞の機能異常が腫瘍形成に伴う組織能力の低下に関わっていることに着眼し、唾液腺上皮細胞の分化・発癌機構の解明を行い、宿主内アスパラギン酸プロテアーゼのカテプシンEを標的分子とする新たな唾液腺悪性腫瘍の診査法を確立することを目的とし、本研究課題を遂行した。
その結果、生体防御システムとの関連が強く示唆されているカテプシンEがマウス唾液腺上皮細胞の分化初期段階で必須の分子であること、また本酵素がWnt関連分子の分子型ならびに局在の制御因子として働くことで、唾液腺上皮細胞の分化に関わっていることをin vitroの系で明らかにし、組織学的解析から、in vivoにおいても同様のメカニズムが存在していることを示唆した。本研究から得られた知見は、プロテアーゼが'分化'と'制癌'とを直接的に結びつけている分子であるという重要な知見を提供するものであった。
