Outline of Research at the Start

これまで我々は、心房細動・心不全といった心臓疾患が発症する遺伝要因や、疾患発症の機序を解明してきました。本研究では、 (1)それぞれの遺伝要因はどの細胞でどのように分子に影響を与えるのか? (2)遺伝・環境要因によって影響を受けた細胞はどのように組織構築に寄与するか? (3)数多くの遺伝・環境要因が分子に与える機能的意義を網羅的に明らかにできるか? (4)上記の分子機序を標的とした治療法を開発して心房細動・心不全を治療できるか? という学術的「問い」を、我々が独自に開発してきた全ゲノム解析・組織シングルセルオミックス解析・変異/遺伝子の網羅的解析システム・創薬スクリーニングを駆使して解明します。

Outline of Annual Research Achievements

我々は、心房細動・心不全において、ゲノム解析で遺伝要因を同定し、組織シングルセル解析で分子挙動や組織構築の機序を解明し、創薬スクリーニングで分子機序を標的とした治療法の開発を進めてきた。本研究では、(1)各遺伝要因はどの細胞でどのように分子に影響を与えるのか?(2)遺伝・環境要因で影響を受けた細胞はどのように組織構築に寄与するか?(3)遺伝・環境要因が分子に与える機能的意義を網羅的に明らかにできるか?(4)分子機序を標的とした治療法を開発して心房細動・心不全を治療できるか?という「問い」に答える研究を推進してきた。2024年度は、心不全で生じる核形態変化とDNA損傷定量から患者の予後を予測する深層学習AIモデルの開発(Hayashi et al. Bioinformatics. 2024)、炎症性心疾患と鑑別が難しい遺伝性心筋症の発見(Nishijo et al. ESC Heart Fail. 2024)、病原性バリアントの重複による心筋症オリゴジェニックモデルの構築(Hiruma et al. Circ Heart Fail. 2024)、体細胞変異で生じるクローン性造血による病態増悪の機序解明(Inoue et al. JACC Basic Transl Sci. 2024)、ビタミンD2によるクロマチンDNA損傷の軽減による心不全治療法(Ito et al. Circulation. 2024)、DNA損傷と関連する非サルコメア遺伝子変異の発見(Dai et al. Circ Heart Fail. 2024)、DNA損傷陽性の老化細胞から分泌されるIGFBP7を標的とした心不全ワクチン治療法の開発(Katoh et al. Circulation. 2024)を進めることができた。
