Outline of Research at the Start

次世代シークエンス技術の発達に伴い、個人・疾患集団間で横断的にオミクス・シークエンス情報を統合し疾患病態を俯瞰する方法論(=統合シークエンス解析)の有用性が注目されている。本研究の目的は、最先端シークエンス技術で構築した免疫アレルギー疾患のオミクス情報を統合し、免疫機構の疾患特異的・動的・経時的な破綻像(=免疫ダイナミクス)を解明し、病態・発症機構を明らかにする点にある。多施設共同・診療科横断的レジストリを通じて多彩な免疫アレルギー疾患の生体試料・臨床情報を収集し、一細胞解析を主軸に多層的オミクス情報を構築する。

Outline of Annual Research Achievements

次世代シークエンス技術の発達は個別のオミクス情報層の免疫アレルギー疾患発症因子の同定を進めたが、個人間・疾患集団間でオミクス・シークエンス情報を統合し疾患病態を俯瞰する方法論(=統合シークエンス解析)は未だ発展途上である。本研究は、最先端シークエンス技術で構築した免疫アレルギー疾患のオミクス情報を統合シークエンス解析で疾患・情報層横断的に統合し、免疫機構の疾患特異的・動的・経時的な破綻像の解明を通じた病態・発症機構の解明を行う。多彩な免疫アレルギー疾患の生体試料・臨床情報を収集し、一細胞解析を主軸とした最先端シークエンス技術を駆使して多層的オミクス情報を構築する。疾患特異的オミクス因子を同定し、独自に新規開発した統合シークエンス解析手法を適用する。令和5年度においては、シングルセルシークエンス解析の対象となる免疫アレルギー疾患を広範囲化し、サンプル数を増加させた上で、更なるシークエンスデータの構築と統合シークエンス解析の解析手法開発を進めた。免疫細胞分化軌道解析など多角的な解析手法の適用に加え、疾患ゲノム情報の一細胞解像度情報への投影による免疫アレルギー疾患の責任免疫細胞集団の同定を行った(例:新型コロナウイルス感染症における自然免疫細胞の重要性)。疾患感受性遺伝子変異が、免疫細胞特異的かつ疾患罹患条件下特異的な遺伝子発現制御効果(cell type and context-specific eQTL効果)を有する例についても、更に多くの感受性変異を見出し、日本人集団におけるカタログ化を進めた。全ゲノムシークエンス情報を活用し、通常のありふれた遺伝子変異だけでなく、これまで見過ごされていた遺伝子変異に着目した解析を進めた。

Strategy for Future Research Activity

引き続き臨床試料の収集と統合シークエンス解析手法の開発を継続し、統合シークエンス解析を通じた疾患層別化病態を規定する免疫ダイナミクス変化の一細胞解像度での解明や、新規疾患治療開発に資する免疫ダイナミクス介入対象の同定を目指した研究を進めていく。
