Outline of Research at the Start

単純ヘルペスウイルスＩ型(HSV-1)のゲノムを遺伝子工学的に改変して、がん細胞で選択的に複製するがん治療用HSV-1を作製できる。研究代表者らは治療域の広い第三世代がん治療用HSV-1（G47Δ）を開発し、世界初の悪性神経膠腫を適応症とした薬事承認を実現した。本研究は臨床実績を踏まえ、次世代に向けたウイルス療法の開発研究を行う。独自の遺伝子組換えHSV-1作製システムを応用し、悪性脳腫瘍の根治を可能にする革新的かつ実用的なウイルス療法開発を狙う。

Outline of Annual Research Achievements

単純ヘルペスウイルスＩ型(HSV-1)のゲノムを遺伝子工学的に改変して、がん細胞で選択的に複製するがん治療用HSV-1を作製できる。研究代表者らは治療域の広い第三世代がん治療用HSV-1（G47Δ）を開発し、世界に先駆けた臨床開発を展開した。2018年には膠芽腫の第Ⅱ相医師主導治験の中間解析で高い治療効果が示されて有効中止となり、2021年6月に世界で初めて悪性神経膠腫を適応症とした国産ウイルス療法製品が承認された。本研究は臨床実績を踏まえ、悪性脳腫瘍の根治を可能にする革新的ウイルス療法の開発研究を行う。３つのアプローチを柱に開発を進めている。BACプラズミドを活用した独自の遺伝子組換えHSV-1作製システムを応用して、G47Δを基本骨格とし、悪性脳腫瘍の治療に応用可能な新規の腫瘍治療用ウイルスを複数作製した。膠芽腫由来がん幹細胞を用いた担がんモデル等で有効性の評価を進めている。世界でウイルス療法の開発が進む中、我々の技術が、悪性脳腫瘍に対する有効性・安全性において抜群に優れていることが治験の結果で明らかになった。技術を更に発展させ、悪性脳腫瘍の根治を可能にする日本独自の、革新的かつ実用的なウイルス療法の基礎開発研究を推進している。

本研究は、世界で初めて悪性脳腫瘍に対するウイルス療法薬の製造販売承認を実現した臨床実績を踏まえ、悪性脳腫瘍の根治を可能にする革新的ウイルス療法の開発研究を行う。３つのアプローチを柱に開発を進めている。BACプラズミドを活用した独自の遺伝子組換えHSV-1作製システムを応用して、G47Δを基本骨格とし、悪性脳腫瘍の治療に応用可能な新規の腫瘍治療用ウイルスを複数作製した。膠芽腫由来がん幹細胞を用いた担がんモデル等で有効性の評価を進めており、研究は順調に進展している。
