もやもや病は難病に指定される進行性の閉塞性脳血管障害であり、進行を止める治療薬はない。我々は患者の8割に共通するRNF213の創始者変異p.R4810Kを同定したが、同変異の浸透率は1%と低く、第2の因子特定が求められてきた。我々は第2の因子としてIL-1βを特定し、RNF213の影響を受けるその下流分子を明らかにしてきた。本研究では、IL-1βとp.R4810K変異がもやもや病の特徴である内膜増殖や線維化を来す分子メカニズムを解明し、その知見を元にマウスモデルを確立することを目指す。
