Outline of Research at the Start

酸素発生型光合成における光誘導水分解・酸素発生反応の機構、及び光エネルギーの吸収・変換を担う各種生物由来光化学系I、光化学系IIと光捕集アンテナタンパク質の超分子複合体等の構造・機能・進化過程を解明する。水分解反応機構の解明では、X線自由電子レーザーを利用したポンプ-プローブ構造解析法を行い、各種変異体の構造・機能解析や理論計算の結果と組み合わせて、O-O結合の形成機構や水の進入経路、プロトンの排出経路を明らかにする。各種超分子複合体の構造解析では、クライオ電子顕微鏡を利用して構造未知の複合体の構造を解明する。得られた結果は、光エネルギーの高効率人工利用にも道を開くものである。

Outline of Annual Research Achievements

水分解反応の機構解析では、時分割ポンプ-プローブシリアルフェムト秒X線結晶構造解析法を用いた、1または2閃光照射し、それによって誘導される光化学系II(PSII)のS1→S2, S2→S3遷移の中間状態の構造変化について、データを取りまとめ、論文としてNature誌に出版した。また、PSIIのプロトンを可視化するため、クライオ電子顕微鏡でPSIIイメージを約10,000イメージ収集し、単粒子構造解析を行った。その結果、1.45Å分解能の電顕マップが得られ、このマップに基づき原子モデルを構築した結果、PSII単量体当たり21,568個の水素(全原子48,461)を構築することに成功した。そして専用計算機を用いて、Mn4CaO5クラスターの高精度量子化学計算(Coupled Cluster(CC)法)を実行し、282原子(QM)モデルにおいてS1,S2,S3状態における様々な電荷、スピン及びプロトン化状態での相対安定性を評価した。
光エネルギーの高効率利用に関しては、シアノバクテリアであるアナベナからPSIと鉄欠乏によって生じる光捕集タンパク質IsiAとの超複合体PSI-IsiAを精製し、その構造を解明した。PSI単量体あたりIsiAが6分子結合し、すでに報告されていたPSI-IsiAの構造とは全く異なった構造であることが判明した。また、紅藻シアニジウムからPSIと光捕集タンパク質LHCIとの超複合体(PSI-LHCI)を精製し、その立体構造を解明した。これまでPSI-LHCIの構造が解明されている近縁の紅色生物のLHCIの分子系統解析を行い、構造解析の結果と組み合わせることによって、紅藻から珪藻やクリプト藻への新たな進化モデルを提案した。

Reason

光化学系II(PSII)における水分解反応の機構解明では、X線自由電子レーザーを利用したポンプ-プローブ法により、1閃光または2閃光照射で、S2またはS3が形成されるまでの過程を、20 nsecーmsecの時間範囲でデータを収集し、それぞれの構造を解析した。その結果、S1→S2, S2→S3の遷移過程で、電子伝達・プロトン移動に伴い、様々な構造変化が観測され、特に2閃光照射後の早い時間で、O6が出現する前にO6*が出現し、それがO6の位置に移動することを見つけ、O-O結合の生成過程に重要な知見を与えた。研究成果はNature誌に公表され、予想以上の成果であると言える。この他、PSIIの超高分解能構造解析、Coupled cluster法を用いた理論化学計算、クライオ電子顕微鏡を用いた光化学系-アンテナ色素タンパク質超分子複合体の構造解析等の項目でも研究は着実に進展したので、当初の計画以上の進展があったと言える。

Strategy for Future Research Activity

光化学系II(PSII)の水分解反応機構解明においては、S状態遷移におけるナノ秒よりも速い時間帯の構造変化を解析する。S1→S2, またはS2→S3の状態遷移において、光照射の強度や微結晶の流速などの条件を検討し、状態遷移が起こる条件を見つけ、該当条件下で、光照射後の速い時間帯で(psecーnsec)の構造変化を解析し、光エネルギーの吸収・伝達に伴う構造変化を明らかにする。また、S3→(S4)→S0の遷移については、結晶のバッファー条件をスクリーニングし、遷移効率の高いバッファー条件を見つけ、(S4)→S0の構造解明のための準備を行う。そしてクライオ電子顕微鏡を用いたPSIIの超高分解能構造解析については、PSIIのクライオ電子顕微鏡イメージをもっと多く取得し、1ムービーの最初の2-3フレームのみを使用して構造解析することで、電子線損傷を最低限に抑えた状態で構造を解析し、プロトンを可視化し、水分解反応やプロトンの放出、水分子の進入に重要なプロトンの存在を明らかにする。得られた構造について、Coupled cluster法を用いた理論化学計算を400原子程度に拡張したモデルについて実行し、各S状態での実験構造を最適化し、反応経路を探索する。
一方、光エネルギーの高効率利用機構の解明では、構造が未解明の緑藻、クリプト藻、珪藻等から光化学系-光捕集アンテナタンパク質超分子複合体を精製し、クライオ電子顕微鏡による構造解析を行い、他の生物由来の光化学系-光捕集アンテナタンパク質複合体構造との比較から、それぞれの生物由来の超分子複合体の特徴、エネルギー伝達経路、及び進化におけるそれぞれの超複合体の変化を明らかにする。
