Outline of Research at the Start

睡眠は動物に普遍的に認められる行動であるが、睡眠覚醒行動を制御する分子基盤は明らかではない。睡眠の不調に苦しむ多くの人の福音となる睡眠介入法の開発には睡眠の根本的理解が必要である。研究代表者らは睡眠制御機構の解明を目指し、世界で類のない哺乳類を用いた睡眠のフォワード・ジェネティクス研究を推進し、ノンレム睡眠を制御する細胞内シグナル系としてSIK3パスウェイを見出した。本研究では、睡眠のフォワード・ジェネティクス研究を推進するとともに、SIK3パスウェイの上流および下流シグナルを同定する。またこれらの分子に着目した研究を通じて、ノンレム睡眠およびレム睡眠制御の分子機構を解明する。

Outline of Annual Research Achievements

睡眠制御パスウェイの解明を目指した睡眠のフォワード・ジェネティクス研究として、顕性スクリーニングとSLP変異のサプレッサースクリーニングを実施した。それぞれのスクリーニングにおいて、睡眠異常家系や睡眠正常化SLP家系を複数樹立した。責任遺伝子変異の同定を行い、睡眠制御機構の解明に向けた分子神経科学的研究を展開した。SIK3パスウェイについては、SIK3のキナーゼ領域をリン酸化酵素LKB1がリン酸化し、SIK3がヒストン脱アセチル化酵素HDAC4およびHDAC5をリン酸化することで、HDAC4/5の細胞内局在が変化し、睡眠促進遺伝子群の転写抑制をさらに抑制するというカスケードが、睡眠を制御する細胞内シグナルの実体であると公表したが、さらにSIK3が視交叉上核において睡眠覚醒のリズムや概日リズム長を制御していることを示すことができた。ノンレム睡眠の量、ノンレム睡眠の質に加えて、暗期初期の急激な覚醒にもSIK3が必須の役割を果たしていることが明らかとなった。人で言えば朝、しっかり目覚てスムーズに日中の活動に移行できることを意味する。SIK3パスウェイが睡眠覚醒の3要素を、異なるニューロン集団を通じて制御していることが2023年度に得られた重要な知見である。SIK3の基質としてHDAC4以外の候補分子が存在することも明らかになった。アデノ随伴ウイルスを用いた操作によって、睡眠制御における役割を検討し、関連を支持る結果を得た。ナトリウムリークチャネルNALCNの変異によるレム睡眠異常の責任部位についてはCreマウスやアデノ随伴ウイルス投与による検討を進めた。NALCNインタラクトームの同定のため近接依存標識NALCNマウスを作製した。ホモ変異マウス脳を用いて蛋白相互作用を検討した。以上のように、ノンレム睡眠からレム睡眠までの神経生物学的な研究を展開し新しい知見を得ている。

Reason

前年度に報告した睡眠の量と質を制御する細胞内シグナル系としてのSIK3パスウェイが、睡眠覚醒のリズムの生成にも必須の役割を果たしていることを公表することができた。抑制性ニューロン特異的なCreマウスや、視交叉上核特異的なCreマウスを用いることで、視交叉上核の非AVP産生ニューロンにおけるSIK3の存在が、明期から暗期に切り替わった直後の急激かつ長く続く覚醒に必須であることが示された。同一の細胞内シグナル伝達系が、異なるニューロン集団を介して睡眠の量、質、さらにリズムを制御していることは予想していなかった知見である。睡眠の進化や起源を考える際の新たな立脚点となるだろう。前年度に報告した睡眠の質と量を制御する領域は大まかなものであることから、Creドライバーマウスやアデノ随伴ウイルスを用いて、責任ニューロン集団の同定を継続している。顕性スクリーニングによって樹立されたレム睡眠が延長する家系で同定された遺伝子変異が、レム睡眠を延長させることを顕性スクリーニングによって見出された。遺伝子改変マウスやアデノ随伴ウイルスを用いて、同定された遺伝子変異がレム睡眠量を変化させることを確認した。SLP変異サプレッサースクリーニングにより覚醒延長の責任遺伝子変異を同定した。SIK3キナーゼ活性に基づくスクリーニングによって同定された、HDAC4以外の基質分子が睡眠覚醒、特にノンレム睡眠中の脳波徐波成分量を変化させることも見出した。以上のように、睡眠制御の分子機構解明につながる多数の発見がなされた。

Strategy for Future Research Activity

睡眠異常の顕性スクリーニングとSLP変異サプレッサースクリーニングは、当初の計画通りに家系の樹立や遺伝子変異同定が進展していることから、計画通りに推進していく。睡眠異常またはSleepy変異によるノンレム睡眠延長に対する正常化作用を示す遺伝子変異の同定と、因果性の確認を進める。睡眠物質とSIK3パスウェイの接続のために、遺伝子改変マウスを用いた検討に加えて、神経活動のイメージングも実施する。SIK3を介したシナプス機能制御解明のため、SIK3のシナプスにおける局在を可視化するとともに、相互作用する蛋白質の同定を続ける。候補蛋白質については、初代神経培養等での機能解析を進めるとともに、遺伝子改変マウス作製やアデノ随伴ウイルスを用いた操作によって、睡眠への影響を明らかにする。単一核トランスクリプトーム解析に基づく仮説ドリブンな研究も展開する。視床下部での睡眠量調節に関わるニューロンタイプやその分子機構を明らかにする。大脳皮質等でのノンレム睡眠中脳波徐波成分制御に関わる分子機構を明らかにするため、皮質層特異的なCreマウスを用いてSIK3の操作を行い、ノンレム睡眠中徐波の変化を評価する。レム睡眠を制御するNALCNの責任部位の同定を終える。
