Outline of Research at the Start

種分化とは、一つの連続的な集団が分かれ、その分化が維持されるに至る一連の過程である。対比的な環境に生息する集団は適応形質が分化し、生態的種分化を始めると考えられる。一方、雑種異常のような内在性の生殖隔離、つまり外部環境に依存しない生殖隔離は、種分化の完成に重要であると考えられている。では、生態的種分化の必然の帰結として、時間が経てば連続的に内在性の雑種異常が進化して種分化は完成に至るのだろうか?あるいは、種分化の完成には何らかの促進因子が重要なのだろうか?本研究では、申請者らが独自に確立してきた種分化段階の異なる野外トゲウオの集団・種のペア(種分化連続体)を利用することで、この問いに答える。

Outline of Annual Research Achievements

10組以上のトゲウオ種間・集団間の交配を行って雑種異常の連鎖マッピングを実施した。イトヨとニホンイトヨの雑種異常に関しては高密度マッピングの結果、X染色体上にかなり限局したピークを見出し1.5Mb以内の領域に原因遺伝子座を絞り込めた。加えて、雑種のバルク及び1細胞RNAシークエンスを実施することで、雑種で発現異常を示す遺伝子のリストアップも行い、複数の候補遺伝子を見出した。また、繁殖期の違いは、同所に生息する種間の重要な交配前隔離の一つであるが、繁殖期の違いに関与する下垂体の甲状腺刺激ホルモンβ鎖2遺伝子(Tshb2)が、既知のホルモン産生細胞群とは異なる新たな下垂体の細胞群で発現していることを1細胞解析によって見出した。また、生息地の分化に関与するFads2遺伝子の周囲に、non-B DNA構造を見出し、これが変異を誘導している可能性が示唆された。さらに、TADなどの3Dゲノムを解析し、TAD境界で逆位が生じやすく、逆位が同所に生息する近縁種間の遺伝子流動の抑制に関与していることを見出した。

Reason

当初想定していなかった新たな発見が複数あった。まず、生息地の分化に関与する遺伝子の周囲を詳細に解析した結果、non-B DNA構造を見出し、これが変異を誘導している可能性が示唆された。つぎに、繁殖期の違いを生み出すTshb2が、下垂体の新規細胞群で発現していることを見出した。さらに、TADなどの3Dゲノムを解析し、TAD境界で逆位が生じやすく、逆位が集団間の遺伝子流動の抑制に関与していることなどを見出した。これらはいずれも当初想定していなかった発見であり、今後の新たな研究の潮流を生み出しうるものであるため。

Strategy for Future Research Activity

上記の遺伝子に加えて、種分化を引き起こす生殖隔離の遺伝子をさらに同定し、集団ゲノム解析を用いて、どのように進化してきたのかを明らかにする。また、どのような条件で種分化が生じやすいのかに関するシミュレーションおよび半野外実験を継続・推進する。これら成果を論文にまとめる。
