Outline of Research at the Start

研究代表者が発見した直鎖状ユビキチン鎖はLUBACリガーゼによって特異的に生成される新規シグナル伝達に寄与する翻訳後修飾である。代表者はこれまで多彩な手法を用いて解析を進め、世界のLUBAC、直鎖状ユビキチン鎖研究をリードしてきた。本研究では予備的知見を踏まえて、LUBACが生成する直鎖状ユビキチン鎖の癌治療への応用、自己免疫疾患、感染症制御、グリコーゲン代謝調節における役割などの研究を推進する。

Outline of Annual Research Achievements

本研究課題では研究代表者が発見し、発展させてきた直鎖状ユビキチン鎖、LUBACの研究の中で、未解明の機能発現メカニズム、LUBACの新たな機能、疾患研究への展開を目指して研究を推進している。3年目である本年度は以下の3項目から研究を推進し、下述の結果を得た。1) 直鎖状ユビキチン鎖生成亢進による自己免疫疾患の発症機構では、昨年度LUBACの機能亢進がヒトおよびマウスの全身性エリテマトーデス(SLE)に寄与することを発表した。その発表論文で、LUBACの機能亢進マウスではI型インターフェロン産生が亢進していることを見出していた。しかしながら、これまでLUBACのI型インターフェロン産生への寄与は報告されていないので、本年度はその分子メカニズムの解析を推進した。2) ヒトでは直鎖状ユビキチン鎖を生成する酵素はLUBACのみであると考えられているが、LUBACは自らのみが生成する直鎖状ユビキチン鎖と特異的に結合するドメイン(NZFドメイン)を2つ有している。そこで、産生する酵素がその生成産物である直鎖状ユビキチン鎖を認識する必要性、役割の解析を推進した。そして、LUBACの直鎖状ユビキチン鎖結合能はLUBACの2つの機能(NF-kB活性化と細胞死抑制)に必須であることを示し、Cell Death Disease誌に報告した。3) HOIL-1L変異のゲノム変異はSLE以外にも異常グリコーゲンの蓄積によるポリグルコサン小体ミオパチーを呈する患者が報告されている。HOIL-1LはLUBACのサブユニットの1つで直鎖状ユビキチン鎖生成に寄与しているが、HOIl-1Lも酵素活性を有する。申請者らはHOIL-1Lの酵素活性がグリコーゲン代謝制御に必須であることを見出していたので、本年度はそのメカニズムの解明を推進した。
