| Outline of Research at the Start |
径の異なる生体材料粒子を作製しマクロファージに貪食・処理させることで、粒子径固有のマクロファージ活性化(炎症促進作用:M1や抗炎症作用:M2)を誘導できることを明らかにする。この基礎的知見を基に、ハイドロゲル内に目的に応じた材質や径の粒子と機能薬剤を組込んだハイブリッドゲルの作製を試み、必要な細胞誘導と適切なマクロファージ活性化によって疾患(特に慢性炎症性疾患やがん)をコントロールする新たな医療材料治療戦略の基盤知見収集を目指す。

| Outline of Final Research Achievements |
Disturbances in macrophage activation form the pathogenesis of various diseases.It was possible to change macrophage activation to inflammatory or anti-inflammatory by administering hydrogels or particles to macrophages. In particular, we have shown that the inflammatory response induced by macrophages that phagocytose particles of a specific diameter inhibits tumor cell growth both in human cell experiments and in therapeutic experiments on implanted tumors in mice.As a secondary result, we also found an interspecies difference in the expression of CD163, a typical marker of anti-inflammatory macrophages, which is not expressed on monocyte-derived macrophages in mice.

| Academic Significance and Societal Importance of the Research Achievements |
マクロファージの代表的機能;1) 異物を貪食・処理すること、2) 環境に応じて炎症性/抗炎症性に可塑的活性化し様々な疾患を増悪すること、が医療材料投与により疾患治癒に最適な活性化状態を作り出し、新規治療戦略として応用できることの基礎的知見を収集することができた。このことは、がんや慢性炎症、自己免疫疾患といった基本治療のみでは根治や疾患コントロールが得られない難治性疾患に対して、横断的な新規治療を提供する可能性を提示できる可能性がある。また本研究を通して代表的なマクロファージ活性化マーカー;CD163の発現に哺乳類間の種差があることを報告し、当該研究領域に重要な知見と警鐘を提供できた。
