Outline of Research at the Start

本研究では、1)膵臓GnRHの産生細胞とその発現機序をアネキシンA5との関連を含めて解明する。2)膵臓におけるGnRH-アネキシンA5のインスリン分泌に対する作用と作用機序を解明する。3 )膵臓GnRHに対する性腺ホルモン(アンドロジェン、エストロジェン)の影響の3点を明らかにする。実験に用いるホルモン測定のための時間分解蛍光免疫測定法、リアルタイムPCR法、in situハイブリダイゼーションを行う。動物、細胞はすでに維持しており、実験手技は確立しているので必要な試薬類を購入してすぐに着手できる。遺伝子変化の網羅的解析のためにRNAseqは委託で行う。

Outline of Final Research Achievements

In type 2 diabetes, the prevalence is generally higher in men than in women, but the origin of this sex difference remains unclear. In this study, we investigated the involvement of locally expressed gonadotropin-releasing hormone (GnRH) in the pancreas in insulin secretion, and examined the role of the gonads in pancreatic endocrine function.The main findings are summarized as follows: 1) GnRH stimulates insulin expression in pancreatic islets. 2) In hpg mice lacking GnRH expression, both annexin A5 and insulin expression are reduced in the islets. 3) Testosterone administration increases insulin expression. These results suggest a functional relationship between the testes and pancreatic islet GnRH, which may contribute to sex differences in pancreatic endocrine function.

Academic Significance and Societal Importance of the Research Achievements

糖尿病は、インスリンの分泌不全や耐性による糖代謝異常で、治療しなければ死にいたる。ヒトの糖尿病は世界的に近年増加傾向にある。2型糖尿病の有病率は男性で高いが、性差がどうして起こるのかよく分かっていない。本研究の成果は、この問題に取り組んだもので、まずイヌにおいても近年糖尿病有病率が増加していること、オスで有病率が高いことを観察した。実験動物を用いて、膵島にGnRHとアネキシンA5が発現しており、インスリンとグルカゴン発現に関わっていること、精巣と機能的に相関していることを明らかにした。膵臓内分泌に関する基礎的知見を得たのみでなく、糖尿病の病理発生解明に資することも期待される。
