Outline of Research at the Start

放射光を用いた円二色性法は,生体膜存在下などの様々な溶媒環境下で,蛋白質の天然と膜結合構造の高精度解析が可能である。本研究では,蛋白質の天然から膜結合構造に至るまでの速度や中間体の形成を観測するため,マイクロ流路を用いた時間分解セルを開発し,サブミリ秒レベルで構造変化を追跡できる円二色性計測システムを構築する。膜内への薬物輸送や膜の安定化などに関わる蛋白質の膜結合構造やその構造の形成過程を精密に解析する。

Outline of Annual Research Achievements

水溶性タンパク質の生体膜相互作用によるダイナミックな構造変化を明らかにすることは,生体膜上で様々な生命機能や物性の発現を理解するために重要である。このため我々は、膜結合タンパク質溶液と生体膜溶液を高効率に混合可能なマイクロ流路時間分解セルを開発し、真空紫外円二色性(VUVCD)分光法に実装し、生体膜相互作用研究のモデルタンパク質であるbetaラクトグロブリンの天然から膜結合構造に至る過程(速度・中間構造)を観測してきた。本年度は、この時間分解測定法(TR-VUVCD)をステロイドホルモンなどの薬物と結合するalpha1-酸性糖タンパク質(AGP)の生体膜との相互作用によるalpha‐beta構造転移の観測に応用した。生体膜にはlysoDMPG由来のミセルとDMPG由来のリポソーム(直径100nm)を使用し、VUVCDスペクトルを波長領域260-180nm、時間領域0.1-120secで測定した。溶媒条件は、AGPが強く膜結合する弱酸性(pH4.5)で行った。これらの相互作用において、模倣膜の曲率がAGPの構造変化の速さと、alphaヘリックス形成量と相関があることが示唆された。相互作用において静電的相互作用・疎水性相互作用のそれぞれが必要であることは判明していたが、曲率の関わる生体膜の疎水性内部の露出の程度が相互作用に大きな影響を与えていることが新しく判明した。また得られたTRスペクトルデータセットより、AGPが膜結合状態へ移行するまでの構造変化経路と静電的・疎水性相互作用の影響の議論が可能となった。

Reason

本研究では、すでに時間分解装置が放射光計測機に実装され、光学システムの評価などが標準試料により完了し、モデルタンパク質の膜相互作用実験にも応用されている。また、本システムを利用することで、膜内への薬物輸送機能を持つとされる膜結合性タンパク質などの構造機能研究が実施されている。さらに、本装置を利用したいくつかの共同研究他も開始したため、当初の計画以上に進展していると判断した。

Strategy for Future Research Activity

今後は、時間分解システムを用いて、alpha1酸性糖蛋白質(AGP)以外の生体膜との相互作用により重要な機能や物性をもつタンパク質の構造変化研究に応用していく。alpha-シヌクレイン(alpha-Syn)は,パーキンソン病の原因となるアミロイド線維を形成する膜結合蛋白質であり,その膜結合構造は線維形成に直接影響する。VUVCD 法よりalpha-Synの膜結合構造(小胞の模倣膜を使用)の二次構造含量・本数・配列を解析し,膜結合する領域や相互作用の種類(静電的・疎水的相互作用)を決定する。またTRCD によりalpha -Syn が膜結合構造に至るまでの速度や中間体の解析を行い,塩(NaCl)の効果を含めて膜相互作用の機構を明らかにする。家族性パーキンソン病を促進するとされる3 種の変異体 (A30P, E46K, A53T)の構造研究も行い,alpha-Syn の膜結合構造,膜相互作用機構,線維形成との関係性の解明を試みる。
