Outline of Research at the Start

本研究では,従来の短いDNA配列解析からは不明瞭であったフタバガキ科樹木の分布域形成の歴史を解明するために,これまでに採取したDNAサンプルを使って,フタバガキ科主要3種の葉緑体ゲノムの全塩基配列を決定する。そして,集団ゲノムデータを集団遺伝学的手法によって解析することで,遺伝的多様性の地理的分布を明らかにする。さらに,研究成果を自然集団の遺伝的多様性を考慮した保全に還元することで,生物多様性保全に関連する施策を支援する。

Outline of Annual Research Achievements

本研究では,従来の短いDNA配列解析からは不明瞭であったフタバガキ科樹木の分布域形成の歴史を解明するために,フタバガキ科主要3種の葉緑体全ゲノムデー タを解析することで,遺伝的多様性の地理的分布を明らかにする。フタバガキ科樹木の葉緑体ゲノム全塩基配列を決定するために,既存のDNAサンプルを再調整し,15Gbの塩基配列を取得した。これらのショットガンシーケンスによるゲノム断片配列データを用いて葉緑体ゲノムのアセンブリをおこなった。

Reason

次世代シーケンス解析用のサンプルを再調整し,葉緑体ゲノム全塩基配列を決定するためのゲノム断片配列データを得ることができた。これによって,葉緑体ゲノム情報に基づいた集団ゲノム解析のために十分な情報を得ることができる。

Strategy for Future Research Activity

対象のフタバガキ科樹木3種複数個体のゲノム配列を決定し,特に地域間の遺伝的差異に関する解析を実施する。
