Outline of Research at the Start

本研究の目的は「末梢炎症反応に脳内マスト細胞が応答し、ミクログリアが活性化される」という仮説を in vivo イメージングにより証明することである。
2 光子顕微鏡を用いた脳内マスト細胞-ミクログリア in vivo 同時イメージング法を開発し、両細胞の解剖学的および機能的解析を行う。両細胞間の相互作用は、それぞれの細胞を薬理学的および分子生物学的に制御することで明らかにする。本研究により「脳内マスト細胞ミクログリア連関」という脳疾患早期の新たな分子病態の解明が期待できる。

Outline of Final Research Achievements

In this study, we investigated the hypothesis that brain-resident mast cells respond to peripheral inflammatory stimuli and subsequently contribute to microglial activation. Using transgenic mice in which mast cells and microglia selectively express fluorescent reporters, we identified a distinct localization of mast cells within specific brain regions. Consistent with our hypothesis, approximately 50% of mast cells were found in direct contact with microglia. These observations not only support the proposed mast cell microglia axis but also suggest that this interaction may play a critical role in regulating brain function. Future studies will further explore the functional significance of this cellular interaction, with a particular focus on the identified brain areas.

Academic Significance and Societal Importance of the Research Achievements

これまで、ミクログリアが脳内唯一の免疫細胞と考えられてきたが、最近ではある種の末梢免疫細胞も脳内に存在し、脳機能に影響を与えることが報告され始めた。実際、脳マスト細胞の存在は既に報告されているが、特定領域での局在は報告されていない。この未知のマスト細胞を解析することで、新しい脳機能の制御機構の解明が期待できる。ひいては、異なる 2 種類の自然免疫細胞、「ミクログリアおよび脳マスト細胞」に着目して研究を進めることで、免疫細胞を切り口とした全く新しい脳科学「免疫脳科学」の創成に繋がる可能性を秘めている。
