Outline of Research at the Start

本研究では、AAVベクターの静脈投与のみで任意の週齢のfloxマウスにおいて細胞種特異的ノックアウト(conditional knockout; cKO)を起こす手法の確立を目指す。申請者が独自に考案したシステムを複数の細胞種特異的プロモーターに適用する事により、マウス遺伝学に新たな技術を提供する事を目指して研究を行う。

Outline of Annual Research Achievements

血液脳関門透過型AAVベクターと、細胞種特異的プロモーターとを組み合わせることにより、floxマウスへの静脈投与のみで、コンディショナルノックアウト(cKO)を行える手法の開発を目指し研究を実施した。
独自に考案したFlpO x dCreシステムとそれぞれの細胞種の特異的なAAVベクターを組み合わせることにより、アストロサイトでは、90%以上の特異性と40%以上のKO効率、ニューロンでは90%程度の特異性と80%以上のKO効率でcKOを起こせることを示した。

Strategy for Future Research Activity

論文ではアストロサイトとニューロンのみで、cKOを起こす手法を検討したが、これ以外の細胞種でも同様の手法が適応可能かどうかを検討していく。
