Outline of Research at the Start

ドーパミンが報酬以外の感覚予測誤差信号を伝達しているかどうか、伝達しているとすればどの脳領域に伝達しているかを調べるため、霊長類脳に蛍光ドーパミンセンサーを遺伝子導入し、感覚連合学習課題を遂行しているときのドーパミン動態を計測する。本研究によりドーパミンの多様な機能を明らかにすることができれば、神経疾患や精神疾患などで見られる感覚鈍麻や感覚過敏を引き起こすメカニズムの解明および症状改善法の開発につながることが期待される。

Outline of Final Research Achievements

Using a fluorescent dopamine sensor, we measured dopamine release in monkeys that exhibited reward-predictive behavior through Pavlovian conditioning. When cue stimuli and rewards were presented, distinct dopamine release patterns were recorded across different regions of the striatum. In the anterior putamen, dopamine signals reflected the difference between predicted and actual rewards (reward prediction error), whereas in the head of the caudate nucleus, dopamine signals corresponded to the expected value of the cue stimulus. Anatomical findings suggest that distinct groups of dopamine neurons project to these respective regions. This study has been published as a research article (Yan, Amita, Nonomura, Inoue, Schultz, Takada. PNAS 2025).

Academic Significance and Societal Importance of the Research Achievements

ドーパミンの報酬予測誤差信号がサルで見つかって以降、ドーパミンの機能について多くのことがわかってきたが、霊長類の脳において報酬予測誤差信号がどこに送られているかはよくわかっていなかった。今回確立した技術を用いることによって、今後、霊長類特有の認知機能や運動機能に関わるドーパミンの神経メカニズム解明が進むと期待される。今回の研究をさらに発展させ、パーキンソン病やレビー小体型認知症といったドーパミンが関与する神経変性疾患の病態解明にも寄与したいと考えている。
