Outline of Research at the Start

本研究課題では、患者の予後に影響を及ぼす大腸がん肝転移指向性の機構を、「がん細胞」と間質細胞による「ニッチ」の両視点から解明し、ニッチの形成に関わるPAI-1を薬剤によって阻害することで大腸がん肝転移を抑制する治療戦略の開発を行う 。具体的には、PAI-1が形成するニッチの形成機構を明らかにし、同機構を阻害する低分子化合物の探索ならびに中和抗体を作製する。これらによってニッチの破綻を誘導し、大腸がんの肝転移を抑制できるかを検討する。

Outline of Annual Research Achievements

Ⅰ.大腸がん-間質細胞の共培養における大腸がん細胞の増殖性変化に対するPAI-1の機能:複数のヒト大腸がん細胞株にPAI-1を加えた時に生じる大腸がん細胞増殖性変化について、PAI-1 ノックアウトマウスを作製して検証したところ、予想通りの結果を得た。
Ⅱ. PAI-1による大腸がん-肝臓間質細胞相互作用を修飾可能な化合物の同定:当研究所が保有している複数の微生物培養液ライブラリーと化合物ライブラリーの中から、PAI-1の阻害を介して大腸がんの増殖性を制御できる微生物培養液および化合物をスクリーニングをすすめ、同培養液に含まれている化合物の同定を行っている。
Ⅲ. PAI-1による大腸がん-肝臓間質細胞相互作用を修飾可能なPAI-1中和抗体の同定:申請者らはこれまで、東北大学の加藤幸成教授らとともに制がん効果を有する様々な抗体を創製してきた。これまでの手法に従い、PAI-1の機能を阻害可能なPAI-1中和抗体の候補を複数得ている。同抗体の効果をin vivoの検討によって確かめた。

Reason

これまでのところ、当初の予定通り順調に進んでいる。試験管内の実験で示されたことを動物実験で確かめるため、ノックアウトマウスの作製を行ったが、無事成功した。同マウスを用いた検討も、我々の予想通りの結果であった。

Strategy for Future Research Activity

これまでのところ、当初の研究計画通りすすんでいる。細胞内シグナル経路の同定のデータは、これまでヒトとマウスの大腸がん細胞のみならず、様々ながん種でも同様の結果が得られた。本年度はこれまで得られた結果の再現性をとり、学会での報告を行うとともに、学術論文への掲載を目指す。
