Outline of Research at the Start

悪性腫瘍の転移メカニズムの解明は、がん治療の大きな課題である。転移過程では、腫瘍細胞の上皮間葉転換(EMT)とその可逆的性質が重要な役割を果たすと考えられている。本研究では、その性質を担う分子基盤を解明するために、可逆的な遺伝子発現調節に関与するエピゲノム制御とエピトランスクリプトーム制御に焦点を当てて解析を行う。各課題は、申請者独自の研究資源と成果をもとに着想したもので、予備的実験結果を既に得ている。可逆性を特徴とするEMTの分子メカニズムを解明し、それを標的とするがん転移抑制戦略の確立に貢献する。

Outline of Final Research Achievements

The metastatic dissemination of malignant tumors critically depends on phenotypic changes in tumor cells, particularly the reversible character of epithelial mesenchymal transition (EMT). To elucidate the molecular mechanisms underlying this plasticity, we focused on epigenomic and epitranscriptomic regulation, which play key roles in reversible gene expression control. Through this analysis, we identified novel epigenomic regulators including histone methyltransferases and histone deacetylases as well as novel epitranscriptomic regulators, such as RNA methyltransferases and methylation reader proteins, that are involved in EMT, invasion, metastasis, and drug resistance of tumor cells. Furthermore, functional perturbation of these factors, either by gene silencing or pharmacological inhibition, resulted in a marked suppression of malignant progression.

Academic Significance and Societal Importance of the Research Achievements

生存率の低下に直結するがんの転移や薬剤耐性に代表される「がんの悪性進展」を制御することは、がんの克服にとって喫緊の課題である。本研究では、悪性進展を誘導するがん細胞の特性(エピゲノム制御、エピトランスクリプトーム制御、幹細胞性など)を分子の相互作用に注目して明らかにしようと試みた。特に、腫瘍の悪性進展の可塑性を担う分子基盤の一端を解明したことによって、それらを標的とする悪性進展の新しい抑制戦略の確立に貢献することができた。
