Outline of Research at the Start

細胞膜上に存在する小さな不動性の突起である一次線毛は細胞内外のシグナル伝達に働き、その構造形成および機能に関する遺伝子の異常が線毛病と言われる一連の疾患を引き起こすことが知られている。しかしながら、一次線毛形成のメカニズムには未解明な点が多く、その全貌は明らかにされていない。そこで本研究では、プロテオーム解析によりこの経路の制御に関わるタンパク質を網羅的に同定し、ゼブラフィッシュを用いたin-vivo解析によりその機能を評価することにより、一次線毛の形成を制御する機構を解明することを目的とした。

Outline of Final Research Achievements

Primary cilia are immotile protrusions on the cell membrane that play a key role in signal transduction between the intracellular and extracellular environments. Genetic abnormalities affecting their formation or function can lead to ciliopathies and various other diseases. As primary cilia are involved in cellular proliferation, differentiation, and maintenance of homeostasis, elucidating their regulatory mechanisms may contribute to the development of novel therapeutic strategies. Previous studies have shown that Trichoplein suppresses ciliogenesis via the ubiquitin proteasome system, but the detailed mechanisms remain unclear. This study aims to elucidate the regulatory mechanisms of primary cilia formation through proteomic analysis and in vivo studies using zebrafish.

Academic Significance and Societal Importance of the Research Achievements

本研究では、一次線毛の形成動態を制御する新規分子を同定し、その生体内機能を明らかにすることで、一次線毛が担う多様な細胞機能の理解を前進させた。特に、組織再生への関与や新たな分子経路の可能性を示した点は、線毛病をはじめとする疾患研究に新たな視点を提供するものである。得られた知見は、従来のシグナル経路とは異なる機構を標的とする創薬戦略の構築にもつながる可能性があり、学術的・社会的に意義の高い成果である。
