Outline of Research at the Start

本研究では、がん細胞で蓄積し、異常なタンパク質の産生の原因となる未成熟転写終結型RNA (ptRNA)のライフサイクルについて、転写による産生、プロセッシング、翻訳、分解まで制御ポイントを細かく分けて詳細に分析する。さらに、大規模データベースを利用したがん種横断的な解析によって、ptRNA発現上昇のリスク因子を探索する。本研究でがんにおけるptRNA発現の脱制御の理解が進めば、がん治療標的のシーズ導出につながることが期待される。

Outline of Final Research Achievements

In cancer cells, aberrant RNAs (ptRNAs) are produced due to premature transcriptional termination within introns. ptRNAs can act either as potentially harmful molecules or as beneficial sources of functional proteins; however, how they are regulated and associated with cancer remains largely unclear. In this study, we analyzed ptRNA expression across various cancer types and revealed their diverse expression profiles, demonstrating their potential utility in classifying cancer patients. We found that ptRNA expression is associated with cellular immaturity and the expression of RNA modification factors. Furthermore, our analysis of ptRNA-derived proteins identified candidate molecules with potential as neoantigens.

Academic Significance and Societal Importance of the Research Achievements

本研究は、がんにおけるRNA異常の中でも未開拓のままである早期転写終結型RNA(ptRNA)に焦点を当て実施した。主要な成果として、がん患者のRNAデータからptRNAを高精度に同定できるパイプラインの構築に成功したことが挙げられる。これを適用することで、がん種ごとに特徴的なptRNAの発現や、同じがん種の患者間におけるptRNA発現の多様性、ptRNAの制御に関わる遺伝子の同定に関する知見が得られることを実証した。さらに、ptRNAから生じる異常タンパク質の評価に関する基盤も構築した。これらの成果をもとに、ptRNA情報に基づくがんの診断や治療標的の探索への発展することが期待される。
