Outline of Research at the Start
孤発性MSI(microsatellite instability)陽性腫瘍は胃癌・大腸癌・子宮体癌に好発する。

Outline of Final Research Achievements
Among gastric cancers, there exists a distinct subtype characterized by abnormally elevated DNA methylation. Notably, many cases within this subtype exhibit tumor development despite retaining a wild-type TP53 tumor suppressor gene. In this study, we identified that aberrant activation of MDM2, a key negative regulator of TP53, plays a significant role in suppressing TP53 function and thereby contributes to tumorigenesis without TP53 mutation. Moreover, we uncovered a novel mechanism by which cells can evade senescence independently of TP53. These findings shed new light on previously unexplained mechanisms of tumorigenesis and are expected to significantly contribute to the development of new therapeutic targets and the advancement of personalized medicine beyond a mutation-centric paradigm.

Academic Significance and Societal Importance of the Research Achievements
