| Outline of Research at the Start |

| Outline of Final Research Achievements |
We revealed that giant cell tumors of bone and chondroblastomas frequently had mutations in H3F3A and H3F3B, respectively, and showed the characteristic expression pattern of the transcriptional factors related to the chondro-osseous differentiation. A certain transcriptional factor gene was methylated in association with H3F3A mutation in giant cell tumors of bone, suggesting that this gene might be responsible for inhibition of the osseoblastic differentiation. In chondrosarcomas often with IDH1/2 mutation, 5-hmC expression level (which represented methylation status) was associated with biological behavior and gene expression pattern. The results suggested that epigenomic alteration may play an important role in the pathogenesis and progression of bone tumors.
