Outline of Research at the Start

トラベクテジンは、肉腫で承認されている数少ない抗がん剤であり、融合遺伝子をもつ肉腫に抗腫瘍効果を示すと言われている。しかし、トラベクテジンの薬効と融合遺伝子の関係性については十分に理解されていない。本研究の目的は、融合遺伝子とトラベクテジンの抗腫瘍効果の関係性を肉腫において明らかにすること、そして肉腫にトラベクテジンが奏効する分子機構を明らかにすることである。患者由来肉腫モデルを用いた感受性試験と、プロテオゲノミクス解析のデータを統合的に解析することで、トラベクテジンの薬効の分子背景を詳細に理解し、奏功性予測バイオマーカーの候補となるタンパク質を同定することを目標とする。

Outline of Final Research Achievements

Approximately one-third of sarcomas harbor characteristic fusion genes, which serve as diagnostic markers and therapeutic targets. Trabectedin is considered particularly effective against fusion-positive sarcomas, yet its molecular mechanism of action remains unclear. To investigate this, the applicant established a panel of 61 sarcoma cell lines derived from patient tumors, representing diverse histological subtypes. Drug sensitivity testing showed that trabectedin had antitumor effects in more than half of the lines, including many fusion-negative cases. This suggests that additional molecular factors contribute to trabectedin responsiveness. To explore this further, protein and RNA expression analyses were performed, leading to the identification of candidate biomarkers associated with drug sensitivity. These findings offer insight into the molecular basis of trabectedin efficacy and support efforts to optimize its clinical application through a biomarker-guided approach.

Academic Significance and Societal Importance of the Research Achievements

肉腫は希少がんであり、症例数が少ないことから市場規模も限られ、製薬企業は新薬開発に慎重である。そのため、治療薬として承認されている抗がん剤の数は非常に少ない。近年、日本ではトラベクテジン、エリブリン、パゾパニブが新たに承認され、一定の治療選択肢が広がっているが、各薬剤の最適な使用法を確立するには、分子レベルでの作用機序や感受性の理解が不可欠である。中でも、トラベクテジンは融合遺伝子陽性肉腫に対して高い効果を示すとされているが、その分子基盤は未解明である。融合遺伝子は種類が多く、機能も異なるため、今後は融合遺伝子と薬剤感受性の関連を体系的に明らかにする研究が求められる。
