Outline of Research at the Start

本研究課題では、ファージ尾部吸着分子をコードする遺伝子群の情報を基に、個々の遺伝子産物を発現・精製し、Clostridioides difficileに対する結合性を調べる。また、本分子の三次元構造解析により、C. difficileへの結合に重要な活性部位を明らかにする。次に、尾部吸着分子のC. difficileレセプター分子を同定し、ファージの宿主認識機構を解明する。得られた情報を基に、C. difficileに得意的なファージをスクリーニングし、CDI発症マウスモデルを用いたin vivo評価を行う。一連の結果を踏まえ、腸内微生物叢を撹乱しない新規CDI治療法の開発に発展させる。

Outline of Final Research Achievements

Clostridioides difficile infection (CDI) is a refractory diarrhea caused by C. difficile. Antimicrobial agents are used for its treatment, but CDI often recurs repeatedly. Therefore, there is an urgent need to establish a new treatment method as an alternative to antimicrobial agents. In this study, we focused on bacteriophages (phage) as a novel treatment for CDI. Phages have the property of targeting and killing only specific bacteria. We generated a gene product involved in the tail adsorption molecule of phage and analyzed its binding to C. difficile. We found that two proteins demonstrated strong binding. We expect to establish a new therapeutic method by clarifying their specificity.

Academic Significance and Societal Importance of the Research Achievements

CDIは、米国において多数の罹患者が報告されており、年間を通じて莫大な医療コストが費やされている。我が国においてもCDIの発生率は、米国と同等あるいはそれ以上であると推定されている。また、米国疾病管理予防センター(CDC)は、C. difficileを「緊急脅威(urgent threat)」に分類している。このような深刻な状況を受け、世界保健機関(WHO)および厚生労働省は、抗菌薬以外のアプローチによる治療法の開発を促進する研究を推進している。本研究の発展により、バクテリオファージの特性を活用したCDIに対する新規治療法の創出につながることが期待される。
