Outline of Research at the Start

細胞の表現型の可塑性は細胞がゲノムの変化なく表現型のみを変化させる能力として理解されている。可塑性は個体発生において重要な役割を果たすことが知られているが、近年、癌細胞、特に癌幹細胞が環境の変化に適応する過程においても注目されている。しかし、腫瘍組織における表現型可塑性の実態が充分に解明されておらず、有効な阻害方法も確立されていない。
本研究では脳腫瘍幹細胞の表現型可塑性を誘導する微小環境因子を広く探索し、明らかにする。さらに、その制御因子を同定し、フェノタイプ・スイッチを予防あるいは阻害することで脳腫瘍幹細胞の治療抵抗性の克服を目指す。

Outline of Final Research Achievements

This study aimed to identify the microenvironmental factors that induce phenotypic plasticity in glioma stem cells (GSCs) and to elucidate their regulatory mechanisms by establishing a multifaceted analytical system. The results of the study showed that interactions between tumor stem cells and immune cells at the invasive front contribute to the induction of plasticity. In addition, cell adhesion molecules and specific transcription factors were identified as candidate regulators of GSC plasticity. Furthermore, novel findings indicated that the microbiota can influence both immune responses and plasticity in brain tumors.

Academic Significance and Societal Importance of the Research Achievements

本研究により、脳腫瘍幹細胞の表現型可塑性が腫瘍浸潤部での免疫細胞との相互作用により誘導され、細胞間接着分子や転写因子がその制御に関与する可能性が示された。さらに、微生物叢が腫瘍免疫や可塑性に影響を与えるという新知見は、悪性脳腫瘍の浸潤メカニズムの理解を深め、治療抵抗性克服に向けた新たな標的の発見につながる可能性がある。
