Outline of Research at the Start

タウ蛋白の疾患特異的シード依存性凝集がストレイン仮説においてどのような役割を果たすかを解明するために、グリア細胞に焦点をあて、アルツハイマー病の危険因子であるApoEを分泌するアストロサイト表面に存在しタウ蛋白をとりこむApoE受容体をCRISPRi遺伝子転写抑制にて同定、マウス海馬にてノックダウンし効果を解析する。細胞内のタウ蛋白とApoE isofrom特異的相互作用はbicisternic vectorを用いる。同時に本機構を応用した高感度cell-free 増幅法 (RT-QUICK)による変性型認知症鑑別血液マーカー開発を行い、ストレイン仮説の本質に迫る。

Outline of Final Research Achievements

A library of 736 existing drugs was screened in a neuroblastoma cell model to suppress seed-dependent tau aggregation in Alzheimer's disease (AD). Four drugs, lansoprazole, calcipotriene, desogesterol, and pentamidine, significantly suppressed AD seed-dependent aggregation in three cases, and lansoprazole, which was also highly effective in an in vitro aggregation system, was ultimately selected. Lansoprazole, administered intranasally to mice for three months, improved motor impairment in mice injected with AD in the basal ganglia. The amount of insoluble tau in the brain and phosphorylated tau pathology were also reduced. Ala substitution analysis suggested that it may bind to the inside of C-type tau.

既存薬ライブラリーから最もタウ凝集を抑制する薬剤を選択したため、アルツハイマー病治療薬として副作用なく、患者の臨床試験に移行でき治療に大きく貢献できる可能性がある。さらに、どのような構造をもつ化合物がタウ凝集を抑制するかを研究する有機化学的なの分子機構解明にも大きな参考となるであろう。
