Outline of Research at the Start

疫学研究から血管リスクがパーキンソン病発症リスクを軽減することが示されており、このような減量の分子メカニズムを明らかにする。そのために、慢性脳虚血をマウスモデルとして作成し、αシヌクレイン代謝に対する影響を解析する。とくに慢性脳虚血に対する反応は、細胞種ごとに異なっており、細胞ごとの解析をエピゲノムレベルで行うこととする。得られたゲノムワイドデータをもとに、慢性脳虚血がどのようにしてαシヌクレイン代謝を変化させているかを理解し、細胞モデルでのより詳細な検討、実際のヒト検体を用いたモデル解析の妥当性評価を合わせて行う。

Outline of Final Research Achievements

This research aims to elucidate the molecular mechanisms underlying the clinical observation, suggested by epidemiological studies, that chronic cerebral hypoperfusion associated with arteriosclerosis reduces the risk of developing Parkinson's disease (PD). The research was developed centering on a mouse model in which a reduction in αSyn pathology is induced by chronic cerebral hypoperfusion, and a cell model system capable of visualizing intracellular αSyn accumulation using bimolecular fluorescence complementation (BiFC) assay. The study comprehensively captured the changes induced by chronic cerebral hypoperfusion in the neuronal epigenome, and subsequently clarified how hypoxia-inducible factor 1α (HIF1α) regulates αSyn metabolism via autophagy.

Academic Significance and Societal Importance of the Research Achievements

本研究は、パーキンソン病(PD)の病態形成において、従来主に悪化要因と考えられてきた動脈硬化リスクが、PDに限っては保護的に作用しうるという疾患特異的な現象の分子基盤を解明する点に高い学術的価値を有する。特に、慢性的な脳血流低下が神経細胞に及ぼす影響を、細胞種特異的なエピゲノム変化という先鋭的な視点から網羅的に解析するアプローチは独創的であり、神経細胞が慢性低酸素というストレス環境に適応していく分子メカニズムの深遠な理解にも貢献する。加えて、低酸素誘導因子HIF1αとαSyn代謝との関連性を明らかにすることは、PDの病態理解に新たな次元を切り開くと期待される。
