Outline of Research at the Start

パーキンソン病やレビー小体型認知症では、異常なαシヌクレイン凝集体の蓄積が病態の主因と考えられている一方で、凝集体が生体脳内で蓄積・伝播する過程で神経障害をもたらす機序には未解明な部分が多い。本研究では、その過剰興奮がパーキンソン病の症状発現に重要な役割を果たすことが示唆されるグルタミン酸作動性神経に着目し、αシヌクレイン伝播モデルマウスを用いて、陽電子断層撮像法や生体核磁気共鳴法等を駆使してαシヌクレイン凝集体伝播に誘導される神経障害メカニズムを明らかにする。さらに、化学遺伝学的手法を用いた神経活動の操作によるαシヌクレイン病態の変化を検証し、病態制御法の開発につなげる。

Outline of Final Research Achievements

How does pathological α-synuclein induce neural dysfunction during its aggregation and propagation through the living brain?
To address this question, we longitudinally evaluated α-synuclein aggregate accumulation using a novel positron emission tomography (PET) ligand capable of visualizing α-synuclein pathology in αSyn propagation model mice , alongside in vivo magnetic resonance spectroscopy (MRS) to monitor changes in brain glutamate levels over time. In parallel, we performed histochemical analyses on ex vivo brains from model mice to investigate the molecular mechanisms underlying the imaging findings. Our results suggest that during the progression of α-synuclein aggregation and propagation, a reduction in glutamate levels occurs in association with volumetric and density alterations of neuronal synaptic boutons, including those of glutamatergic neurons.

Academic Significance and Societal Importance of the Research Achievements

パーキンソン病やレビー小体型認知症では、異常なαSyn凝集体の蓄積が病態の主因と考えられている一方で、凝集体が生体脳内で蓄積・伝播する過程で神経障害をもたらす機序には未解明な部分が多い。本研究で得られた成果は、αSyn凝集体蓄積・伝播の過程でのグルタミン酸作動性ニューロンの障害を支持する。ヒト病態を良好に再現した疾患モデル動物において、マウスとヒトの双方で利用可能な生体イメージング手法を駆使して得られた本研究の成果は、今後、αSyn凝集体の蓄積を特徴とする疾患のさらなる病態解明と、疾患修飾法開発に向けたトランスレータブルな研究開発への貢献が見込まれる。
