Outline of Research at the Start

自閉症スペクトラム症(ASD)は社会的コミュニケーションの障害を発達早期から示す。ASDの発症機序は遺伝因を含めた器質的基盤による脳の機能障害によるとされているが、未だ明らかとなっていない。今回血漿中に存在する細胞外小胞エクソソームの変化から、ASDの病態に迫れないかと考えた。ASD患者血漿から1)神経由来のエクソソームを抽出しASDの病態との関連を検討する。そして、脳血液関門を通過するとされるエクソソームが、実際に脳-末梢の双方向性の移動が生じていることについて、ASD患者からの血漿エクソソームを解析するだけでなく、2)ASD患者の血漿エクソソームが神経系に影響することを動物実験で確認する。

Outline of Final Research Achievements

The elevated miR-20a-5p in neural-derived exosomes was confirmed in autism spectrum disorder and Alzheimer's disease. Experiments using animals suggest that it is involved in the antidepressant effect. In fact, IRF1 gene expression and miR-20a-5p expression were elevated in depression model mice and postmortem brains of depressed humans. Considering that miR-20-5p has an antidepressant effect and suppressively regulates IRF1 gene expression, it was suggested that miR-20a-5p is elevated in a compensatory manner to improve the pathology of depression, including neuroinflammation.

Academic Significance and Societal Importance of the Research Achievements

患者血液から採取される神経由来エクソソーム内の変化が脳病態を反映する可能性が示唆されたことから、精神神経疾患を通常の採血から判定できる可能性がある。現在用いられている診断基準は症状、またその持続期間から診断する目的で使用されるが、血液から客観的な情報が得られれば診断の精度、画一性が上昇することが期待されるため、社会的意義は大きいと考える。
