To investigate the effects of cellular senescence on dementia and depression in the elderly, a large-scale cohort study was conducted in Nakayama Town, Ehime Prefecture, and over 1,500 blood samples were collected. Risk factors for dementia and mild cognitive impairment (MCI) were identified, including diabetes, hypertension, and elevated high-sensitivity C-reactive protein (hs-CRP). In Alzheimer's disease (AD), increased inflammation associated with cellular senescence, impaired mitochondrial function, and reduced lipid metabolism were confirmed. Additionally, we established methods for measuring microRNA in blood and analyzing gene expression, focusing on cellular senescence, inflammation, and autophagy, thereby shedding light on some aspects of the pathophysiology of dementia and depression. Currently, we are analyzing the relationship between cellular aging and AD from multiple perspectives, including proteomics, metabolomics, and lipidomics.
