本研究では、DNP-MRIおよび炎症で産生される活性酸素との反応性を有するレドックス感受性DNP造影剤を用いた炎症性筋炎の高感度可視化法を開発し、筋炎モデルマウスでの本法の有用性を実証することを目的とする。

| Outline of Final Research Achievements |
In this study, we performed in vivo redox imaging using dynamic nuclear polarization MRI (DNP-MRI) to noninvasively evaluate the inflammatory state of skeletal muscle in mdx mice, a widely used model for Duchenne muscular dystrophy (DMD), one of the representative inflammatory diseases. Using the signal decay rate of the redox probe CmP, we analyzed the redox status of skeletal muscle in mdx mice. At all ages, the decay rate of CmP radicals was significantly faster in mdx mice compared to normal mice, suggesting the presence of a chronic inflammatory environment. Notably, increased ROS production and macrophage accumulation were found to promote the reduction of CmP radicals, indicating that DNP-MRI may serve as a valuable noninvasive method for assessing skeletal muscle inflammation in DMD.

| Academic Significance and Societal Importance of the Research Achievements |
デュシェンヌ型筋ジストロフィー(DMD)は小児期に発症し進行性に悪化する難治性筋疾患であり、早期診断と病態進行の評価が極めて重要である。本研究で開発・検証されたDNP-MRIによるレドックスイメージング技術は、筋組織の炎症状態を非侵襲かつ高感度に可視化できる新たな手法として、患者への負担を軽減しつつ、治療効果のモニタリングや新規治療法の評価に貢献する可能性がある。これは医療現場への応用が期待される社会的に意義の高い技術であるといえる。
